چکیده

Biomass charcoal powder (BCP) was used as a carrier for immobilization of α-chymotrypsin through adsorption. BCP was derived from plant biomass wastes such as dumped bamboos by oxygen-free pyrolysis at low temperatures and grinding with a jet mill. The activity of adsorbed α-chymotrypsin was strongly dependent upon the kind of BCP. The thermal denaturation curve of adsorbed α-chymotrypsin was shifted to high temperature, compared to that of free one. When α-chymotrypsin adsorbed onto BCP of bamboos was incubated at 45 ̊C, the half-life of adsorbed α-chymotrypsin was 2.6 times greater than that of free one. After incubation at 45 ̊C, the remaining activity of adsorbed α-chymotrypsin markedly depended on the kind of BCP.

Effect of Adsorption Condition on Thermal Stability of Proteins Adsorbed onto Biomass Charcoal Powder

Authors have found out that bamboo charcoal powder (BCP), which is prepared from bamboo wastes by pyrolysis at low temperatures, is very useful as a carrier for the thermal stabilization of proteins. Hen egg white lysozyme (HEWL) and α-chymotrypsin (CT) were effectively adsorbed onto BCP. The thermal stability of BCP-adsorbed HEWL was strongly dependent upon the adsorption conditions such as so...

The activity of glucosyltransferase adsorbed onto saliva-coated hydroxyapatite.

This study aimed to determine physical and kinetic properties of glucosyltransferase (GTF) adsorbed onto hydroxyapatite (HA) surfaces. For development of a solid-phase enzyme assay, 4.0-mg samples of washed HA powder were exposed to centrifuged whole saliva (WSHA) or buffer, and subsequently exposed to a GTF solution. The activities of GTF adsorbed to HA and that remaining in solution were meas...

In Vitro Adsorption Studies of Paracetamol to Activated Charcoal Capsule, Powder and Suspension

Introduction: Adsorption capacity of activated charcoal to paracetamol was determined at simulated gastric environment, pH 3.4 and simulated intestinal environment pH 7.2. Three formulations of activated charcoal; powder, capsule and suspension were tested. Paracetamol was derived from tablet to simulate in vivo intoxication conditions. Methods: Activated charcoal and paracetamol were mixed at ...

Biosorption of chromium(VI) and arsenic(V) onto methylated yeast biomass.

Yeast biomass was methylated in a 0.1 M HCl methyl alcohol solution at room temperature and the methylated yeast (MeYE) was applied to the adsorptive separation of Cr(VI) and As(V) anions from aqueous solutions. At near-neutral pH, while Cr(VI) and As(V) anions were scarcely adsorbed onto unmethylated yeast biomass, the amounts adsorbed increased with increasing methylation degree. The amount o...

A comparative study of the conformational stabilities of trypsin and -chymotrypsin

A comparative study was performed on the conformational stabilities of trypsin and α-chymotrypsin. At 45oC, trypsin was most stable at pH 3, while the highest stability of αchymotrypsin was observed at pH 5. With both ester and amide substrates, trypsin displayed activation at pH 3. In the case of α-chymotrypsin, activation was detected at pH 5 only with the amide substrate. The time curves of ...
